Campylobacter spp. are major foodborne pathogens, with C. jejuni and C. coli being the most common species associated with human disease. Poultry and livestock serve as important reservoirs, and secretion system–related genes such as flaA and virB11 play critical roles in pathogenicity. In this cross-sectional study, 392 samples from chickens, cattle, sheep/goats, and water sources in Behbahan (Iran) were analyzed. Isolation and identification of Campylobacter were performed using standard microaerophilic culture methods, phenotypic tests, and PCR targeting the 16S rRNA gene. The virulence genes flaA and virB11 were detected by specific PCR assays. Statistical analyses used chi-square and Fisher’s exact tests. Campylobacter spp. were detected in 12.8% (50/392) of samples. Chickens showed the highest prevalence (20.3%), followed by water (10.7%), cattle (5.7%), and sheep/goats (4.9%). Molecular identification revealed 72% C. jejuni and 28% C. coli. The flaA gene was present in 66% of isolates and was significantly associated with species (P < 0.05), being more common in C. jejuni. No isolate carried the virB11 gene. Poultry represents the main reservoir for potentially virulent Campylobacter strains in the region. The high prevalence of flaA highlights the virulence potential of circulating isolates. Enhanced hygiene and surveillance measures are recommended.
