The efficiency and stability of anaerobic digestion technology depend on complex microbial communities and their activities. Microbial diversity plays a crucial role in the performance of natural and engineered ecosystems. Over the past decade, significant progress has been made in identifying key groups of microorganisms that influence anaerobic digestion processes. Recently developed next-generation sequencing technologies can be used for a comprehensive description of the microbial community in a digester. In the present study, this technique was used to identify and analyze the microbial structure present in an anaerobic digester of urban wastewater sludge. Samples were taken from the effluent sludge flow of an urban wastewater sludge anaerobic digester at different time intervals to perform molecular identification tests using next-generation sequencing technology. The samples then underwent whole-genome sequencing analysis to obtain findings regarding the genetic composition and diversity of the microbial community (species type and abundance). Alpha diversity indices showed that the studied anaerobic digester had a rich, diverse, and relatively balanced microbial community. Bacterial community analysis revealed that Proteobacteria and Firmicutes were the dominant bacterial phyla in the anaerobic digester, while the archaeal community consisted of acetoclastic, hydrogenotrophic, and methylotrophic methanogens. Methanothrix had the highest abundance in the studied digester. The appropriate diversity of bacteria in the studied digester indicates good stability and optimal capacity for anaerobic digestion processes, as well as functional redundancy in the microbial community, which can increase the system's resistance to stresses and operational changes. The higher abundance of Methanothrix suggests that the acetoclastic methanogenesis pathway is likely dominant in the digester. The higher abundance of Methanothrix also indicates that there are no functional disturbances in the digester. Overall, the analysis of the microbial structure showed that the studied digester is in an optimal condition in terms of digestion process stability and consistent methane production.
